Barnsley captain Brian Howard scored a later winner to send Premier League giants Liverpool tumbling out of the FA Cup following the 2-1 result.
Howard lashed home in injury-time to leave the Kop in bemused silence.
Liverpool had been on top for long spells and Dirk Kuyt put them ahead in the first half, before Stephen Foster headed a 56th-minute equaliser for the Championship club.
