Barcelona's pint-sized magician, Messi, is bidding to become the first player to win the award in four successive years, to seal the Argentine's status as the greatest player of his generation.
The 25-year-old is the overwhelming favourite to see off the challenge of Barca team-mate Iniesta, and great Real Madrid rival Cristiano Ronaldo for the prestigious prize.
It will crown another landmark year for Messi, who is the top scorer in La Liga this season with 25 goals, having also broken Gerd Mueller's 40-year record for the most goals in a calendar year, finishing 2012 with an astonishing 91 goals in all competitions.
Portugal captain Ronaldo admits he would love to win his first Ballon d'Or since 2008, but added it would not be the end of the world if the prize went elsewhere.
Iniesta, meanwhile, looks set to miss out despite playing a pivotal role not only for Barcelona over the past 12 months, but also in Spain's record-breaking triumph at Euro 2012 in Poland and Ukraine.
Messi and former French star, UEFA president Michel Platini, are the only players to win three straight Ballon d'Or awards, while Dutch stars Johan Cruyff and Marco Van Basten have also won three accolades apiece.
All three nominees will be in action one last time before Monday night's ceremony in Zurich, as Real Madrid take on Real Sociedad, and Barcelona face Espanyol in La Liga on Sunday evening.
