The Black Cats pulled off a surprise 2-1 victory at the Stadium of Light in the first leg, with a goal from on loan Liverpool striker Fabio Borini and an own goal from veteran Ryan Giggs.
Nemanja Vidic scored for the away side but will not feature in tonight's game after he was shown a red card in United's 3-1 loss to Chelsea.
Sunderland have only lost once since the turn of the year, and are unbeaten in four games in all competitions.
But Gus Poyet's side have only won twice away from home all season and will have to improve when they play United if they are going to make it through to the final.
Manchester United have lost four of the last five games in all competitions only picking up a win against Swansea in the league and Sunderland may fancy their chances when they go to Old Trafford as it isn't the fortress it used to be.
The Red Devils have injury doubts over Patrice Evra, Wayne Rooney and Robin van Persie. Sunderland will be missing Keiren Westwood and Carlos Cuellar, with Steven Fletcher a doubt due to an Achilles injury.
The Black Cats have only won two of the last 25 meetings with United, the first being in the 2000-01 season and the second this year in the first leg.
Manchester United have not lost consecutive League Cup games since September 2007.
The winner will play Manchester City at Wembley after they demolished West Ham 9-0 on aggregate.
